Resume:

2005-2008, Ph.D thesis: Construction of Recombinant Pseudorabies Virus Expressing Rabies Virus Glycoprotein and Research of Immunity Test. We have used it to construct a recombinant virus, rPRV/eGFP/rgp, expressing the rabies virus glycoprotein using Pseudorabies virus, which has been licensed for use as a live vaccine in pigs and possesses an excellent safety and efficacy record. This recombinant virus has been shown to be safe for dogs by oral and intramuscular routes of inoculation and was demonstrated to induce immune responses against both pseudorabies and rabies in dogs after a single oral dose of 2 × 107.0 plaque forming units (PFU). Neutralizing antibody titers against rabies reached >0.5 IU/ml and 1:64—1:128 against pseudorabies by 5 weeks post-vaccination in all dogs, indicating that the pseudorabies virus vector infected dogs and replicated in vivo, and that the rabies virus glycoprotein had been expressed and an effective immune response elicited. Anti-body titers were maintained for over 6 months. This suggests that pseudorabies virus could be an effective live vector for recombinant rabies oral vaccination.  Results were published in Vaccine.

I have 10 years of research experience in Parasitology and 4 years for Rabies and Canine Influenza Virus. I graduated in Jun 2008, and worked in department of preventive veterinary, South China Agricultural University. In May, 2011, we went to the lab charged in associated professor Cunlei Su to study technology related to T.gondii, University of Tennessee.

My research focus has been predominantly molecular and immunological parasitology. My scientific output to date consists of 2 books (in Chinese); more than 78 papers in English in well-regarded international journals (SCI cited) and more than 30 papers in refereed national journals. I have brought 3 research students for PhD degree, 9 research students for MSc degree and 27 research students for Bachelor to completion, and I am now supervising 2 associate professors.

Mastering of experimental techniques:

Molecular biology: DNA and RNA isolation and purification, DNA cloning, library construction and screening, Suppression subtractive hybridization (SSH), Southern blotting, Northern blotting, agarose gel electrophoresis,  PCR; long PCR; Cycle DNA sequencing, Random amplified polymorphic DNA (RAPD)-PCR, PCR-linked restriction fragment length polymorphism (PCR-RFLP), PCR-based single strand conformation polymorphism (PCR-SSCP), Simple sequence repeat-PCR (SSP-PCR), Rapid amplification of cDNA ends (RACE), RNA interference (RNAi), microRNA technology, etc.

Immunology: Preparation and purification of parasite antigens for application in immunoassay and immunization trials; establishment, standardization and quality control of immunoassay; establishments of ELISA, Dot-ELISA, immuohistochemical staining techniques, Immunofluorescence assay (IFA), immunoblot (Western Blot), immunization protocols, SDS-PAGE and isoelectric focusing electrophoresis (IEF); vaccine development; FACS analysis of blood lymphocyte phenotypes; in vitro cultivation of blood/mesenteric lymphocytes for mitogenic responses, antigenic responses and cytokine production.

Research Interests:

Functional genomics, Immunology of parasites; molecular cloning and expression of functional genes of parasites; vaccine development; genetics and molecular diagnosis of parasitic diseases; PCR-based technology in parasitology; epidemiology and control strategies for parasitic infections; Proteomics and microRNA of T.gondii.

But, the current research directions: the invasion mechanism of Toxoplasma gondii breaking through the blood-brain barrier; and the mechanism of Toxoplasma gondii causing reproductive dysfunction.
